Civil War veteran Dan Evans (Christian Bale) lost his leg during the war, and now he and his wife Alice (Gretchen Mol) and sons William (Logan Lerman) and Mark (Benjamin Petry) are about to lose their Arizona ranch to the expanding Southern Pacific Railroad. Dan sees a way out of his financial problems when he is offered $200 to transport stagecoach robber Ben Wade (Russell Crowe) to Contention City where he is to catch the 3:10 train on the way to his hanging at the Yuma prison. Also serving with Dan on the assignment are Tucker (Kevin Durand) who burned down Ben's barn, railroad boss Grayson Butterfield (Dallas Roberts), Pinkerton agent Byron McElroy (Peter Fonda), and Dan's teenage son William. As Dan and Ben match wits along the trek, they also encounter problems with Apaches and Ben's gang–led by Charlie Prince (Ben Foster)–on what turns out to be a dangerous journey to catch the 3:10 to Yuma. Virgil and Everett are setting out to find witnesses to Bragg's crimes when a piano-playing widow, Allie French (Renee Zellweger), arrives in town with plans of her own for stealing Virgil's heart. Soon, the friendship of Everett and Virgil is tested when both men fall for Allie, but they must unite if they are to achieve their mission of keeping the witnesses alive and sending Bragg to court.
