  Artie DeVanzo (Artie Lange) is an unemployed 35-year-old loser who lives with his mother (Laurie Metcalf). Artie's slow-pitch softball team–sponsored by Ed's Bar & Swill–regularly loses to the team of Artie's nemesis, Dennis Manganelli (Anthony DeSando), from high school days. Enough fights between the teams have broken out that Artie's team is in jeopardy of being forced out of the league–unless they can pull their act together and end the team's perennial losing streak.

  Real estate agent Peter Klaven (Paul Rudd) is engaged to Zooey (Rashida Jones) and now has the problem of finding a Best Man for their wedding. Peter does not shine at male bonding and has no male friends. Hoping to help Peter, Zooey urges him to reach out and try to find male friends, hoping he will find a Best Man in the process. Peter picks up on Zooey's suggestion and meets Sydney Fife (Jason Segel). They hit it off–maybe too well because now Peter and Sydney become inseparable, which, of course, causes a problem with Peter's relationship with Zooey.

  When lazy pothead Dale Denton (Seth Rogen) stops by the apartment of his dealer Saul Silver (James Franco), Saul offers him a rare strain of marijuana, Pineapple Express. Dale and Saul smoke for a few minutes, and then Dale, who is a process server, heads out to serve his subpoena on drug lord Ted Jones (Gary Cole). While waiting to approach Jones, Dale witnesses crooked cop Carol (Rosie Perez) and Jones kill a man. Dale panics and flees the scene, leaving behind his still-smoldering joint. Jones finds the joint, gets help from middle man Red (Danny McBride), and tracks it back to Saul and Dean. Now, hit men appear on the scene, and Dale and Saul try to escape while bullets and body parts fly through the air.

Danny mentors geeky teenager Augie Farks (Christopher Mintz-Plasse) who loves playing a Dungeon & Dragons type game, "Lare." Wheeler is assigned to work with Ronnie (Bobb'e J.   With their high-school graduation just two weeks away, good friends Seth (Jonah Hill) and Evan (Michael Cera) are looking forward to attending Jules' (Emma Stone) party. In fact, Seth and Evan have taken on the job of supplying the booze for the big event. So, with nerdy friend Fogell (Christopher Mintz-Plasse)– who has managed to get his hands on a fake Hawaiian drivers' license–in tow, the trio goes shopping. They send Fogell into the store, and, wouldn't you know it, the liquor store is robbed while he is inside. Fogell is knocked unconscious, and Seth and Evan ditch him. Now, Fogell ends up in the custody of two inept police officers Slater (Bill Hader) and Michaels (Seth Rogen) and spends the night cruising. Meanwhile, Seth and Evan head off with a total stranger, Francis (Joe Lo Truglio), who tells them he will help them get the party's liquor. And the adventures continue.
