Stand-up comedian George Simmons (Adam Sandler) learns that he suffers from a rare form of leukemia and has less than one year to live. George meets young Ira Wright (Seth Rogen) who dreams of becoming a comedian, and he offers Ira the job of working as his assistant and performing as his opening act. Meanwhile, George tries to reunite with his former girlfriend Laura (Leslie Mann), but her husband Clarke (Eric Bana) does not agree to the plan. Now, George learns that his disease has gone into remission, and he reevaluates his lifestyle and decides to find what will really give his life meaning.
